Rett syndrome is a unique neurodevelopmental disorder that is first noticed in infancy and primarily affects girls, but can be rarely seen in boys. Rett syndrome can go undiagnosed or misdiagnosed, making it difficult to determine the disorder’s true frequency in the general population, and is mostly misdiagnosed as autism, cerebral palsy, or global developmental delay. Its diagnosis is based on the identification of characteristic symptoms, a detailed patient history, and a thorough clinical evaluation. Further, molecular genetic testing can detect the presence of mutations of the MECP2 gene and confirm the clinical diagnosis of Rett syndrome.

Treatment for the disorder is symptomatic — focusing on the management of symptoms — and supportive, requiring a multidisciplinary approach. Antiepileptic drugs can be used to help control seizures; A class of antidepressants, known as selective serotonin reuptake inhibitors (SSRIs) can be used for the treatment of anxiety; Proton pump inhibitors (PPIs) or H2 blockers can help reduce reflux; Laxatives can be used to treat constipation. The US FDA has recently approved DAYBUE (trofinetide) to treat Rett syndrome in adult and pediatric patients two years of age and older.
