Several limitations have been observed in current in vitro antibody display methods. On one hand, these methods cannot provide naturally paired heavy chain and light chain of antibodies, which could potentially lead to the generation of antibody molecules that have suboptimal solubility or other biophysical characteristics. As a result, antibodies derived from these technologies most likely do not represent the naturally cognate heavy and light chains. On the other hand, the antibody derived from these methods generally have the moderate affinity to the targets and require further affinity maturation before used as potential therapeutics. Thereby, phage libraries are restricted to about 1010 to 1011 total diversity by the efficiency of bacterial transformation as well as by the practical limit of phage particle density of about 1013/mL.
